Cytogenetic Analysis: This is a hypotetraploid human cell line. The modal chromosome number is 87, occurring at 50%, with higher ploidies at 1.6%. Eight markers [der(1)t(1;9) (p31.2;q11), 6p+, del(6) (p21.1), t(12p14q) and four others] were found in all cells. Of these markers, six (including all four listed above) were consistently paired and two others were single. Normal N6 and N9 were absent and N7 had six copies per cell. There were only two X chromosomes per cell.

Comments: The WM-115 cell line was derived from the primary tumor.

Another cell line derived from a metastasis in the same patient also is available (ATCC CRL-1676).

The cells should be incubated at 34C to 35C.

Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated Eagle's Minimum Essential Medium, Catalog No. 30-2003. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.

Temperature: 34.0°C; (Max. 35.0 °C, Min. 34.0°C)

Atmosphere: air, 95%; carbon dioxide (CO2), 5%

Subculturing: Protocol:

Remove and discard culture medium.

Briefly rinse the cell layer with 0.25% (w/v) Trypsin- 0.03% EDTA solution to remove all traces of serum that contains trypsin inhibitor.

Add 2.0 to 3.0 ml of Trypsin-EDTA solution to flask and observe cells under an inverted microscope until cell layer is dispersed (usually within 5 to 15 minutes).

Note: WM-115細(xì)胞To avoid clumping do not agitate the cells by hitting or shaking the flask while waiting for the cells to detach. Cells that are difficult to detach may be placed at 34 to 35?C to facilitate dispersal.

Add 6.0 to 8.0 ml of complete growth medium and aspirate cells by gently pipetting.

Add appropriate aliquots of the cell suspension to new culture vessels.

Incubate cultures at 34 to 35?C.

Subcultivation Ratio: A subcultivation ratio of 1:3 to 1:6 is recommended
